Biopreparaadi Bactomix 5 toime ristõieliste nuutritekitaja (Plasmodiophora brassicae) arengu takistamisel

Abstract
Ristõieliste nuutrit põhjustav mullapatogeen Plasmodiophora brassicae on mullas võimeline säilima ligikaudu 20 aastat. Tänasel päeval efektiivsed tõrjemeetodid nakkuse kontrollimiseks puuduvad. Alternatiivse tõrjemeetodina on uuritud mullas esinevaid baktereid, mis on in vitro katsetes tõestanud mullapatogeene allasuruvat toimet. Käesoleva bakalaureusetöö eesmärgiks on teha kindlaks biopreparaadi Bactomix 5 potentsiaal ristõieliste nuutritekitaja (P. brassicae) arengu takistamisel, samuti rakendada ja hinnata kvantitatiivse PCR (qPCR) metoodika sobivust P. brassicae puhkeeoste hulga määramiseks mullast. Uurimusest selgus, et mulla töötlemisel kasutatud biopreparaat Bactomix 5 mõju ristõieliste nuutrit põhjustava patogeeni P. brassicae alla surumisel ei ilmnenud ja patogeeni kogus mullas ei vähenenud. Seejuures oli qPCR metoodika sobiv ja usaldusväärne viis P. brassicae püsieoste kvantitatiivsel määramisel mullast. P. brassicae hulga suur varieerumine erinevate pottide muldade seas näitab metoodika täpsustamise vajadust proovide maksimaalse homogeniseerimise näol. Edaspidiseid katseid on soovitatav läbi viia kontrollitud toimeainet sisaldavate bioloogiliste preparaatidega ristiõieliste kultuuride piires.
 
Plasmodiophora brassicae is an economically significant soil borne pathogen causing club root disease in Brassica species resulting in considerable yield losses among crucifer crops. The pathogen is difficult to manage since its resting spores are viable in soil for many years and the cultural practices of the disease are limited. Biological control has proven to be an appropriate alternative in fighting plant pathogens. Some species of bacteria have the ability to supress diseases, exhibiting activities against plant pathogens. The aim of this study was to analyse the effect of a certain bacterial product Bactomix 5 on the development of clubroot and it’s potential pathogen supressive activity and assess the suitability of real-time PCR method for detection and quantification of P. brassicae resting spores in the soil. The results showed no significant effect of Bactomix 5 on the development on P. brassicae. On the other hand, qPCR proved to be rapid and sensitive method for quantifying P. brassicae DNA. For more detailed conclusions, further research based on testing different biocontrol products on P. brassicae is needed. Products containing only one active agent rather than a mixture of multiple substances is advised.
