| dc.description.abstract | Töö uurib importpuidus ja –taimedes olevaid patogeene ning patogeenide tahtmatut introdutseerimist uuele alale või regiooni. Antud küsimust Eesti mastaabis ei ole varem uuritud ja väheseid töid leidub selle teema kohta ka teistes keeltes. Töö eesmärgiks on: 1) dokumenteerida patogeenide invasiooni imporditud puittaimede istikutega ning puidust pakkematerjaliga; 2) määrata tahtmatult introdutseeritavate patogeenide liigid, s.h ka EPPO A2 nimistu karantiinsed liigid; 3) dokumenteerida patogeenide päritolumaad. Analüüsiti 20 proovi imporditud puitpakenditelt, 93 importtaime proovi Hollandist, Saksamaalt, Rootsist ja Jaapanist. Uurimuses kasutati molekulaarseid praimereid Diplodia sapinea, Phytophthora spp., Lecanosticta acicola ja Dothistroma septosporum, kandseene hõimkonna ja seente tuvastamiseks üldiselt. Üks proov puidust õnnestus sekveneerida ja liik tuvastada. Kokku 113 analüüsitud proovist 97 proovi oli positiivsed seeneriigi taksonite suhtes, s.o 85 %. Analüüsitud proovidest tuvastati patogeen perekond ja liik 6 juhul, s.o 5%. Kolmel juhul määrati D. sapinea, ühel juhul D. septosporum, ühel juhul Phytophthora perekond ning ühest proovist kandseen Pachylepyrium carbonicola. Töö raames avastati maailmas esmakordselt serbia kuuselt (Picea omorika) D. septosporum. | est |
| dc.description.abstract | This thesis investigates the pathogens in imported timber and plants and their introduction into other regions. This problem has not been investigated in Estonia yet and similar subject matter from other countries is also scarce. The goals of the thesis were: 1) documenting inadvertent introduction of pathogens via plant and wooden packing material; 2) determine the species of introduced fungi including EPPO A2 list quarantine species; 3) documenting the invasive pathogens country of origin. All samples were analysed moleculary and in this study species-specific PCR primers were used for identifying fungi. In the study was used Diplodia sapinea, Lecanosticta acicola, Dothistroma septosporum, Phytophthora spp., Basidiomycotaspecific-primers. Also fungi universal primer was used in the investigation. Furthermore one wood sample was sequenced. Altogether 113 samples were in the research. A pathogen was found in 6 samples. In three samples D. sapinea was detected and in three different samples D. septosporum, Phytophthora spp and also fungus from Basidiomycota were detected. The last positive sample was sequencedand determined Pachylepyrium carbonicola. This study is the first one in the world to detect Dothistroma septosporum on Picea omorika in the world. The thesis shows what kind of pathogens are being inadvertently introduced into Estonia via imported wooden packaging material and plants. | eng |
