Abstrakt
Magistritöös kasutatud materjal koguti institutsionaalse uurimistoetuse projekti „Piimalehmade sigimine ja tervis.“ (IUT8-1) raames. Eesmärgiks oli uurida kolm nädalat enne ja kolm nädalat pärast poegimist erinevas toitumuses eesti holsteini tõugu lehmade süsivesikute ja lipiidide ainevahetust iseloomustavate vere metaboliitide dünaamikat vastusena glükoosi infusioonile. Valimisse võeti 44 multipaarset eesti holsteini lehma, kes jagati vastavalt toitumushindele kolme rühma: alamäärases toitumuses (≤ 3,0), optimaalses toitumuses (3,25-3,5) ja ülemäärases toitumuses (≥ 3,75). Intravenoosse glükoosi tolerantsi testi ajal võeti vereproovid sõltuvalt glükoosi infusioonist aegadel: -5., 5., 10., 20., 30., 40., 50., 60. minutil. Vereproovidest määrati glükoosi, esterifitseerimata rasvhapete, β-hüdroksübutüraadi ning triglütseriidide kontsentratsioon. Glükoosi infusioon tõi endaga kaasa lühiajalised muutused loomade ainevahetuslikes seisundites, mille dünaamikad rühmade vahel olid sarnased, kuid erinesid absoluutväärtustes. Lehmade ainvahetus poegimisjärgselt oli intensiivsem, mida näitas glükoosi kiirem kadu verest. Samuti leiti, et ainevahetuse seisund on seotud varulipiidide hulgaga enne poegimist.
This Master`s thesis is a part of the institutional research grant „Fertility and health in dairy cattle“(IUT8-1).The objective of this Master`s thesis was to evaluate the response of metabolites characterising carbohydrate and lipid metabolism to glucose infusion in Estonian Holstein dairy cows. Intravenous glucose tolerance test (GTT) was performed three weeks before the expected parturition and three weeks after the actual parturition. Multiparous Estonian Holstein dairy cows (n=44) were divided according to body condition score (BCS) into three groups: under conditioned (BCS ≤ 3,0), optimal conditioned (BCS 3,25-3,5) and over conditioned (BCS ≥ 3,75). During GTT blood samples were collected at -5, 5, 10, 20, 30, 40, 50 and 60 min relative to administration of glucose. Glucose, nonestrified fatty acids (NEFA), β-hydroxybutyrate (BHB) and triacylglucerol (TG) concentrations were analyzed from blood samples. The concentration dynamics of glucose, BHB, NEFA and TG were comparable between three groups, but differed in values. The results confirmed the hypothesis that the metabolic status and glucose tolerance depend on the amount of cows’ fat reserves in dry period.
