Abstrakt
Diplodia sapinea on Eestis kiiresti leviv seenpatogeen. Maailmas on D. sapinea laialt levinud
seenhaigus ning esineb elavates puu kudedes isegi endofüüdina. Töö eesmärgiks oli invasiivse
D. sapinea leviku analüüs ning selle seene latentse faasi olemasolu kindlakstegemine Eestis,
sporulatsiooni uurimine ning kahe praimeripaari võrdlusanalüüs D. sapinea varajase
diagnostika eesmärgil. Töös kasutati konventsionaalse PCR analüüsi DiSapi ning Bot ja Dp
DNA praimerite paare. Antud töö tulemusena D. sapinea’d endofüüdina hariliku männi
proovidest ei leitud. D. sapinea puhaskultuure analüüsides näitasid Bot ja Dp praimerid 48
positiivset tulemust ja 16 negatiivset tulemust ning DiSapi praimeritega olid kõik 84 proovi
positiivsed. Seega on D. sapinea diagnostikaks parim Eestis disainitud praimeripaar DiSapi
ning kasutatav edaspidistes teaduslikes ja rakendusteaduslikes töödes. Patogeeni
sporulatsiooni uuringust selgus, et Eestis on D. sapinea eoste leviku maksimum novembri
kuus.
D. sapinea is rapitly spreading pathogen in Estonia. D. sapinea is wildly spread in the world and can be in a living tree tissue as an endophyte. The aim of this thesis was to determine the range of invasive D. sapinea spread analysis in Estonia, to examine D. sapinea in latent phase in Scots pine shoots, to investigate the sporulation of D. sapinea in Scots pine and to compare two species-specific primers for earlier diagnostics. DiSapi and Bot, Dp primers were used for conventiual PCR analysis. D. sapinea wasn’t in the collected healthy Scots pine shoots. In the species-specific primers comparison, there were 68 positive samples and 16 negative samples with Bot, Dp primers and all the 84 samples were positive with DiSapi primers. The research revealed that DiSapi, which is designed in Estonia is the most effective primer pair to diagnose D.sapinea. The peak of D.sapinea sporulation was at the beginning of November according to the findings.
