Kirje Ainevahetuslikud vastused glükoosi infusioonile erinevas toitumuses eesti holsteini lehmadel(2015) Karis, Priit; Jaakson, Hanno (juhendaja); Ling, Katri (juhendaja)Magistritöös kasutatud materjal koguti institutsionaalse uurimistoetuse projekti „Piimalehmade sigimine ja tervis.“ (IUT8-1) raames. Eesmärgiks oli uurida kolm nädalat enne ja kolm nädalat pärast poegimist erinevas toitumuses eesti holsteini tõugu lehmade süsivesikute ja lipiidide ainevahetust iseloomustavate vere metaboliitide dünaamikat vastusena glükoosi infusioonile. Valimisse võeti 44 multipaarset eesti holsteini lehma, kes jagati vastavalt toitumushindele kolme rühma: alamäärases toitumuses (≤ 3,0), optimaalses toitumuses (3,25-3,5) ja ülemäärases toitumuses (≥ 3,75). Intravenoosse glükoosi tolerantsi testi ajal võeti vereproovid sõltuvalt glükoosi infusioonist aegadel: -5., 5., 10., 20., 30., 40., 50., 60. minutil. Vereproovidest määrati glükoosi, esterifitseerimata rasvhapete, β-hüdroksübutüraadi ning triglütseriidide kontsentratsioon. Glükoosi infusioon tõi endaga kaasa lühiajalised muutused loomade ainevahetuslikes seisundites, mille dünaamikad rühmade vahel olid sarnased, kuid erinesid absoluutväärtustes. Lehmade ainvahetus poegimisjärgselt oli intensiivsem, mida näitas glükoosi kiirem kadu verest. Multiparous Estonian Holstein cows (n = 42) were assigned to three experimental groups on the basis of BCS 28 days before expected calving (d –28) as follows: BCS ≤ 3.0 (2.25–3.00; thin (T), n = 14); BCS = 3.25–3.5 (optimal (O), n = 14); BCS ≥ 3.75 (3.75–4.50; over-conditioned (OC), n = 14). Blood samples were taken between d –21 and d 42 in relation to calving, milk production data were collected throughout lactation. The OC cows' adaptation to the demands of lactation was the worst based on the comparison of dynamics of blood parameters between BCS groups. They had the most unbalanced metabolism and used more stored lipids compared to T and O cows. Loomade heaolu tähtsustamisest lähtuvalt on parandatud veiste pidamistingimusi. Söötmiskorraldus on teadlik ja eesmärgipärane. Parandatud on põhisöötade (eeskätt silo) kvaliteeti. Söödaratsioonide koostamisel minnakse sügavuti, need on täpsed ja võtavad arvesse üha enam parameetreid. Seda kõike tehakse selleks, et tagada kõrge aretusväärtusega piimakarja igakülgne toitefaktoritega varustatus, hea tervis ja jõudlus ning pikaealisus. Eestis on senistes piimaveistega läbi viidud uuringutes enam tähelepanu pööratud laktatsiooniperioodi lehmade söötmisega seotud küsimustele, kinnislehmade söötmisega seonduv on aga jäänud tahaplaanile. Sellest tulenevalt seati uuringu lähteülesandeks välja töötada Torma POÜ-le Eesti tingimustesse sobiv ja kaasaegsetel seisukohtadel põhinev piimakarja kinnisperioodi söötmisstrateegia, mille tulemusena paraneb piimalehmade tervis, karjas püsimine, heaolu ja produktiivsus. Selleks tuli kõigepealt välja selgitada puhkeperioodi söödaratsiooni energiakontsentratioon, mille korral lehmade toitumushinne kinni jätmisest ettesöötmise alguseni (ca 21 päeva enne loodetavat poegimist) ei muutu. Most of the disease events occur during that period; this highlights the need for deeper understanding of cows’ adaption to lactation to increase the welfare of cows and profitability of farm. Insulin is a pancreatic hormone that has a key role in energy metabolism, in general, it promotes storage of energy within organism. Insulin resistance is a state when insulin causes less than normal response in target tissues. It causes intensive lipolysis, which is a risk factor for the occurrence of numerous metabolic diseases. This doctoral thesis elaborates on the relationships of the volume of adipose tissue during dry period with lipolysis, insulin resistance, and the expression of genes and concentration of proteins in adipose tissue that regulate glucose and lipid metabolism. Four weeks before expected calving multiparous Estonian Holstein dairy cows (n=42) were grouped into three according to their body condition score. Three weeks before and after calving intravenous glucose tolerance test and biopsy from subcutaneous adipose tissue were performed. After the formation of experimental groups, blood samples were taken weekly and body condition was assessed fortnightly. Milk samples were taken and zootechnical data registered until the end of the lactation. The energy corrected milk production of over-conditioned cows was higher than that of optimal or thin cows, but the higher production was achieved at the expense of intensive use of energy stored in adipose tissue. Cows that were more resistant to insulin action before calving were prone to intensive lipolysis at the beginning of lactation, which was related to lower expression of lipogenic genes and lower concentration of glucose transporter 4 in subcutaneous adipose tissue. Results clearly show that excessive volume of adipose tissue increases metabolic load and the risk of metabolic diseases.
